Validation of liquid chromatography mass spectrometry (LC-MS) methods

Self test 2.1

1. Separation factor α characterizes the ability of the system to resolve chromatographic peaks.

2. Separation factor α characterizes the selectivity of the chromatographic process but completely disregards the efficiency of the process.

3. Peak resolution RS characterizes the ability of the system to resolve chromatographic peaks.

4. Peak resolution RS characterizes the selectivity of the chromatographic process but completely disregards the efficiency of the process.

5. In order to achieve good resolution between peaks both selectivity and efficiency of the chromatographic process are important.

6. Please find the resolution (RS) between the peaks of acetophenone and toluene. You may print the chromatogram for measuring the peak widths. Note, that for this calculation retention times and peak widths can be expressed in arbitrary units (minutes, millimeters, inches etc) as long as the same unit of measure is used for all quantities. Therefore, if you print the chromatogram and measure retention times and peak widths with ruler, there is no need to convert them to minutes.

Alternatively you can press to the arrow on the right of the chromatogram to have the chromatogram with the numerical data.

Please present the result with two significant digits.

Rs=

7. Can the two peaks of the chromatogram in question 6 be considered resolved?

8. Does the chromatogram in question 6 imply any other selectivity issue apart from partial overlap between the peaks of acetophenone and toluene?
