A2. Synthetic protein tags and regulatory circuits

Group 3 with assistance from molecular design group 1 aims to develop a universal toolbox of sisu phosphorylationbased protein tags for fine tuning of target protein expression and localization in engineered producer strains (10). We will test the system by modulation of the expression of the central carbon metabolism enzymes for optimized and efficient production of biobased chemicals. The toolbox of easily tunable sisuphosphorylation based protein tags will be further developed into more complex synthetic signaling circuits that will involve metabolite sensors connected to different feedback and feedforward loops for dynamic control of producer cell metabolism. The task will be started in yeast and then extended to other organisms involved in the current application (Groups 2, 4 and 5).
